To clarify how the extraction agent pH value influences the protein and
polysaccharides release, we selected the following extraction agents: 5.0 sodium
hydroxide solution, 5.0% hydrochloride and isotonic 0.9% solution of sodium chloride.
The extractions were conducted at the temperature of 50 ± 2 ºС, which facilitates the
allergenic material extraction and does not lead to the protein denaturation [6, 7, 9].
For conducting the allergenic material extraction usually the following ratio was
used: 1 g of raw materials : 100 ml (more rarely 50 ml or 150 ml) of extracting agent,
because at this ratio the raw materials: extracting agent the allergenic extract formation
is possible, which may be used during the skin tests without additional concentration
or dilution. For further research we chose 100 ml of the extracting agent.
To prevent the contamination all manipulations were conducted in the laminar
box with the maintenance of aseptic conditions. The dry C.albicans fungus biomass
was obtained and milled in the mortar, weighted 1 gr and in parallel the protein and
polysaccharides were extracted in 100 ml of corresponding extraction agents. One part
was extracted with 5.0% solution of sodium hydroxide, the other – 5.0% solution of
hydrochloride acid and the third one - with isotonic 0.9 % solution of sodium chloride
at the temperature of 50 ± 2 ºС. The water-bath was used for the maintenance of the
temperature 50±2 ºС. Considering the information of references on the technologies
of obtainment the extract from the C.albicans fungus biomass, the period of studying
the kinetics of the protein and polysaccharides extraction does not exceed two days.
Thus, the extraction lasted from 1 to 24 hours with further determination of
protein and polysaccharides in the obtained extracts. To bring the obtained allergenic
extracts to the neutral value of рН 7,2 ± 0,2, they were neutralized by the corresponding
chemical substances. In case of extracting using the solution of sodium hydroxide, the
obtained extract was treated by 0.5% solution of hydrochloride acid, and in case of
extracting with the solution of hydrochloride acid, the obtained extract was treated with
0.5 of solution of sodium hydroxide. The preliminary filtration was conducted on the
membrane filters with the diameter of 0.8 µm as well as the sterilization filtration on
the membrane filters with the diameter of pores 0.22 µm.
To define the speed of the allergenic material accumulation in extracts, the
biochemical analysis was conducted of the composition of extracts obtained: Every
hour the protein content was determined according to State Pharmacopoeia of Ukraine
using the Lowry's method as well as the content of polysaccharides using the phenol-
sulfur method.
According to the results obtained, it was determined that the greatest amount of
protein is extracted for the shortest period of time from the biomass of C. albicans
fungus in case of administration of 5.0% of sodium hydroxide solution.
