ultrasound disintegration on the equipment UZUU-21 at the frequency of 22 khz,
intensity 5 W/cm2 and at the temperature of 25± 2 °С. The temperature of 25 ± 2 °С
was constantly controlled during the biofeedstock ultrasonication and maintained by
adding the cold water into the surrounding container. As the apparatus УЗУУ-21
cannot operate for a long time without break, every 30 min. the process was stopped
for 5 min., or, if necessary, for a longer period. Only the operating time was taken as
an extraction time. The protein content was determined every hour. The extracts
obtained were treated with 5.0% solution of hydrochloride acid, gradually bringing the
environment to the pH value of 7.2 ± 0.2. The prior filtration on the membrane filters
was conducted with the pores diameter of 0.8 µm as well as the sterilization filtration
on the membrane filters with the pores diameters 0.22 µm.
The extraction lasted from 1 to 24 hours with further determination of protein
content in the obtained extracts.
According to the results obtained, it was found that the period of time necessary
for the protein release from C. albicans biological mass for the physical and chemical
method, the extraction with 5.0% solution of sodium hydroxide at the temperature of
25 ± 2 ºС in the combination with the ultrasound, was the shortest.
The determination of content of the obtained extracts of C. albicans fungus was
aimed at providing the biochemical characteristics and comparing the biochemical
composition of extracts obtained with the help of different methods (table 2).

The results of studying the composition of the obtained extracts of C. albicans
fungus testify to the fact that in the extract obtained during the treatment with 5.0%
solution of sodium hydroxide in combination with the ultrasound the greatest amount
of protein and polysaccharides was found for the shortest period of time.
To determine the monosaccharide composition of polysaccharides of the obtained
extracts the paper chromatography was conducted.
The Polysaccharides of the extraction with 5.0% solution of sodium hydroxide in
combination with the ultrasound and polysaccharides of the extraction with 5.0 %
solution of sodium hydroxide with mixing were represented by the monosaccharides:
mannose, glucose, tracks of ribose and two non-identified monosaccharides.
